SRP54 / YPR088C Phenotype

Summary
SRP54/YPR088C is an essential gene in reference strain S288C; deletion is inviable in S288C and reported inviable in XJ24-24a, though some assays in XJ24-24a note severe growth impairment rather than absolute lethality. Conditional and partial-loss-of-function alleles reveal broad defects: a temperature-sensitive allele shows abnormal chromosome/plasmid maintenance and reduced killer toxin resistance; dominant-negative SRP54 reduces protein accumulation; reduction-of-function variants cause increased cold sensitivity, decreased vegetative growth, and lower competitive fitness. Repressible alleles lead to increased cell size and decreased protein and mitochondrial transport, accompanied by slower exponential growth and abnormal protein accumulation. Other phenotypes include increased mitochondrial rho− mutation frequency and decreased transposable element transposition in null backgrounds, while nuclear export and RNA accumulation have been reported as normal in some assays, with mixed effects on protein accumulation. Heterozygous diploids are haploinsufficient.
