Summary
BDP1 encodes a subunit of the TFIIIB complex, which is an initiation factor for RNA polymerase III (Pol III). Pol III is responsible for high-level transcription of tRNAs, most small nuclear RNAs, and 5S rRNA. Two other subunits of TFIIIB are TATA-binding protein (TBD), encoded by SPT15, and Brf1p. The recruitment of TFIIIB to DNA upstream of the transcription initiation sites depends on another Pol III initiation factor, TFIIIC, which recognizes and binds intragenic Pol III promoters. The assembled TFIIIB-DNA complex distorts DNA over the start site and recruits Pol III, as a result facilitating transcription initiation as well as multiple rounds of re-initiation at the promoter. Transcription by Pol III is negatively regulated by Maf1p, which under growth-limiting conditions becomes dephosphorylated in the cytoplasm, translocates to the nucleus and binds to the TFIIIB complex, preventing Pol III transcription.
