Summary
RRN6, RRN7, and RRN11 encode protein subunits of the RNA polymerase I (Pol I) core factor (CF) complex, which aids in the recruitment of Pol I to the rDNA promoter. CF enhances polymerase binding to the promoter and properly positions the transcription start site. CF is recruited to the Pol I preinitiation complex after binding of the upstream activating factor (UAF) to the upstream activating sequence (UAS) and recruitment of TATA-binding protein (TBP; Spt15p). TBP cannot activate Pol I transcription without CF. UAF, CF, and TBP form an initiation scaffold at the promoter for Rrn3p-bound Pol I. CF recruits Rrn3p-bound Pol I to rDNA promoters through interactions between the CF subunit Rrn6p and Rrn3p. Rrn7p and Rrn11p interact weakly with TBP, while Rrn6p interacts strongly with TBP. Rrn7p also interacts with the UAF subunit Rrn9p, which itself interacts with TBP, providing an additional protein contact between CF and TBP.
