Summary
TOA1 and TOA2 encode the large and small subunits of the TFIIA basal transcription initiation factor required for RNA polymerase II (RNAPII) transcription initiation. TFIIA acts as a coactivator by interacting with activators and other components of the basal initiation machinery to mediate promoter recognition and unwinding. TFIIA, other basal initiation factors, RNAPII, and promoter DNA together comprise the preintiaion complex (PIC). TFIIA associates with, and stabilizes, the TBP-TATA complex, and functions to counteract the repressive effects of negative cofactors such as NC2. The high sequence and structural conservation of TFIIA from yeast to human underlines the significance of TFIIA in fundamental aspects of eukaryotic transcription.
