Summary
ROX3 (also called MED19) encodes a component of the head module of the Mediator transcriptional regulation complex. The Mediator complex is highly conserved from yeast to human, and is a central player in the transcriptional regulation of protein-coding genes, being required for nearly all transcription by RNA polymerase II (RNAPII). Mediator bridges activators and the general transcription machinery. The core Mediator complex contains three distinct head, middle, and tail modules. Other subunits of the head module are encoded by MED6, MED8, MED11, SRB2/MED20, SRB4/MED17, SRB5/MED18, and SRB6/MED22. The head module binds weakly to a minimal preinitiation complex (PIC) consisting of RNAPII, general initiation factors TFIIF, TFIIB and TBP, and promoter DNA to stimulate basal transcription. Mediator interacts physically with diverse DNA-binding transactivators to communicate transcription signals to RNAPII and general initiation factors to turn transcription on or off. Mediator also has important functions in post-initiation steps, such as recruiting and coordinating the activities of RNAPII elongation factors.
