Summary
RRN5, RRN9, RRN10, and UAF30 encode subunits of the upstream activating sequence (UAS)-binding upstream activating factor (UAF), along with histones H3 and H4. The UAF is a general transcription initiation factor complex that aids in the recruitment of RNA polymerase I (Pol I) to the ribosomal DNA (rDNA) promoter. Pol I preinitiation complex (PIC) formation begins with UAF binding to the UAS, followed by recruitment of TATA-binding protein (TBP; Spt15p), the Pol I core factor (CF) complex, and Rrn3p-bound Pol I. The histones likely provide much of the UAF's affinity for DNA, while the Uaf30p subunit is thought to provide sequence specificity. UAF subunit Rrn9p facilitates PIC formation through interactions with CF subunit Rrn7p and TBP. No UAF analog has been identified in mammalian cells.
