Summary
NDD1 encodes a transcriptional coactivator that regulates progression of the cell cycle in concert with the transcription factors Mcm1p and Fkh2p. Its targets are genes expressed in the G2/M phase of the cell cycle, including cyclin genes CLB1 and CLB2, the CDC5 polo-like kinase gene, the CDC20 component of the APC ubiquitin ligase, and the transcription factor genes ACE2 and SWI5. Fkh2p and Mcm1p bind together at target promoters and repress transcription in the absence of Ndd1p. NDD1 transcription is activated during S phase by the Hcm1p transcription factor, and once Ndd1p is expressed, it binds to Fkh2p at target promoters. This binding is enhanced by phosphorylation by the Cdc28p/Clb2p and Cdc5p kinases, and the binding of phosphorylated Ndd1p to Fkh2p causes transcriptional activation of target genes. If DNA damage occurs, the Rad53p and Dun1p kinases phosphorylate Ndd1p to downregulate it, preventing its binding to Fkh2p at promoters and thus arresting the cell cycle. At the end of M phase, Ndd1p is targeted for degradation by the APC-Cdh1p ubiquitin ligase.
