Summary Paragraph [TOP] [NEXT] Help
SUMMARY PARAGRAPH for NUP49/YGL172W for NUP49
NUP49 encodes an essential nuclear pore protein (1). Transport of macromolecules between the nucleus and the cytoplasm of eukaryotic cells occurs through the nuclear pore complex (NPC), a large macromolecular complex that spans the nuclear envelope (reviewed in 2, 3, 4, 5). The structure of the vertebrate NPC has been studied extensively; recent reviews include 6, 7, 8, and 9. The yeast NPC shares several features with the vertebrate NPC, despite being smaller and less elaborate (10, 11). Many yeast nuclear pore proteins, or nucleoporins, have been identified by a variety of genetic approaches (reviewed in 2, 3, 12, 13, 14). Nup49p is one of a group of nucleoporins that contain multiple repeats of the amino acids GLFG (15, 2); however, the Nup49p repeats are not essential for NPC function (16). Nup49p and Nup57p are very similar, and may have arisen by gene duplication (2). Similar nucleoporins are also found in rat and S. pombe (2). Nup49p is found in a subcomplex of the NPC, often called the Nsp1p subcomplex, that also contains Nsp1p, Nup57p, and Nic96p, (2, 17, 18). This complex has been detected at both the nuclear and cytoplasmic periphery of the NPC central channel (19). Further, Nup49, Nup57p, and the C-terminus of Nsp1p have been expressed in E. coli, and form a complex in vitro (17). Mutations in NUP49 cause defects in nuclear protein import and nuclear RNA export (20, 2); nup49 mutations are also synthetically lethal with mutations in other nucleoporin genes (15, 2).
