Abstract: Monothiol glutaredoxins are localized in a variety of cellular compartments where they are involved in diverse functions. However the subcellular localization and specific function of yeast monothiol Grx3 are unclear. An antibody for native Grx3 was produced to accurately determine the localization of Grx3 in the cell since the use of bulky tags can alter protein localization. The presence of native Grx3 in the cytosol and crude mitochondrial fractions was determined by western blot analysis. Further purification of mitochondria indicated Grx3 localization into a membrane-bound organelle. Additionally, using site-directed mutagenesis, we have determined that the GRX3 start site is Met36 confirming that the molecular mass of Grx3 in vivo is 28 kDa.
