Dietrich F (2011) Personal Communication, An updated S. cerevisiae reference genome: Resequencing of S288C using NextGen sequencing technology. ()
Abstract: The "S288C 2010" version of the Saccharomyces cerevisiae reference sequence was determined at Duke University, from an individual AB972 yeast colony. The strain AB972 was obtained from Maynard Olson (Riles et al Genetics. 1993, 134: 81-150). The sequencing was carried out using 36 base Illumina HiSeq sequencing. The analysis of this data was such that reads were mapped to the previous reference genome and any regions or repetitive sequence were excluded. This resequencing only covers regions of unique sequence in the genome, about 90% of the whole genome. Polymorphism calls were checked by hand to exclude a number of dubious calls. About 10% of the genome contains repetitive sequences. The repetitive regions were excluded from this analysis, including microsatellites, Ty, delta, telomeric region sequences, tRNA sequences, other miscellaneous repeats and a small number of high GC rich regions where the sequence coverage was low.
