NUP170 encodes a nuclear pore protein (1, 11). Transport of macromolecules between the nucleus and the cytoplasm of eukaryotic cells occurs through the nuclear pore complex (NPC), a large macromolecular complex that spans the nuclear envelope (reviewed in 11, 12, 13, 14). The structure of the vertebrate NPC has been studied extensively; recent reviews include 15, 16, 17, and 18. The yeast NPC shares several features with the vertebrate NPC, despite being smaller and less elaborate (19, 20). Many yeast nuclear pore proteins, or nucleoporins, have been identified by a variety of genetic approaches (reviewed in 11, 12, 21, 22, 23). Nup170p is one of the most abundant yeast nucleoporins (1). Nup170p interacts physically with other abundant yeast nucleoporins, Nic96p, Pom152p, Nup157p, and Nup188p (24, 11). These abundant nucleoporins may form the structural core of the NPC (11). Also, Nup170p forms a complex with the nucleoporins Nup53p and Asm4p (25). The complex is found on both the nuclear and cytoplasmic faces of the NPC, and associates with Pse1p, a member of a protein family implicated in nuclear protein import. Nup157p and Nup170p are very similar to each other and to a mammalian nucleoporin, Nup155 (1, 3, 11). NUP170 is not essential, but nup170 mutations are synthetically lethal with mutations in several other nucleoporin genes (26, 11).
