PET100 encodes an integral protein of the mitochondrial inner membrane protein that is required for assembly of cytochrome c oxidase (1, 2). Pet100p acts as a chaperone to specifically mediate the assembly of a Cox7p-Cox8p-Cox9p subcomplex (subunits VII, VIII, and VIIa respectively) into the cytochrome c oxidase holoenzyme (1). Protein-protein interactions are detected between Pet100p and this subcomplex, but not between Pet100p and the holoenzyme (2).
The pet100 null mutant displays a respiratory growth defect and lacks cytochrome c oxidase activity although all of the subunits are synthesized (3). Two cytochrome c oxidase subcomplexes accumulate in a pet100 null mutant: one containing Cox7p, Cox8p, and Cox9p (subunits VII, VIII, and VIIa) and another containing Cox5Ap and Cox6p (subunits Va and VI) (2).
