SUMMARY PARAGRAPH for RHO1

RHO1 encodes an essential small GTPase in the Rho/Rac subfamily of Ras-like GTPases (5). Other S. cerevisiae Rho GTPases are encoded by RHO2, RHO3, RHO4, RHO5, and CDC42 (6). Like other eukaryotic Rho-type GTPases, Rho1p is involved in the establishment of cell polarity, playing a role in the reorganization of the actin cytoskeleton (6). Rho1p regulates protein kinase C (encoded by PKC1) and the cell wall synthesizing enzyme 1,3-beta-glucan synthase (encoded by FKS1 and GSC2) (3, 4). Rho1p is localized to the plasma membrane at sites of growth such as incipient bud sites, bud tips, and the bud neck during cytokinesis (5). The localization of Rho1p is aberrent in sla1 null mutant cells (7). At the restrictive temperature, some temperature-sensitive rho1 mutants arrest with small buds (5); other temperature-sensitive rho1 alleles cause arrest as unbudded cells (2). The GTPase activity of Rho1p is regulated by the GTPase activating proteins (GAPs) Bem2p, Sac7p, Bag2p, and Rdi1p, and the guanine nucleotide exchange factors (GEFs) Rom1p and Rom2p (6).
