RHO2 encodes a non-essential small GTPase in the Rho/Rac subfamily of Ras-like GTPases (1). Other S. cerevisiae Rho GTPases are encoded by RHO1, RHO3, RHO4, RHO5, and CDC42 (2). Rho2p may play a role in the establishment of cell polarity or in microtubule assembly (2). Deletion of RHO2 causes increased sensitivity to the microtubule depolymerizing drug benomyl (3). Overexpression of RHO2 suppresses the temperature sensitivity of cik1 or kar3 deletions; CIK1 and KAR3 encode microtubule-associated proteins (3). The GTPase activity of Rho2p is positively regulated by the GTPase activating protein (GAP) Bem2p, and negatively regulated by the guanine nucleotide exchange factors (GEFs) Rom1p and Rom2p (2).
